Product Description

Synovial joints are unique functional elements of the body and provide the ability for locomotion and for physical interaction with the environment[1,2]. The inner lining of the joint consists of the synovium, also called the synovial membrane, a thin layer located between the joint capsule and the joint cavity. Fibroblast-like synoviocytes (FLS) are mesenchymal origin cells located inside the synovial membrane. FLS display many characteristics common with fibroblasts, such as expression of several types of collagens and protein vimentin, a part of cytoskeletal filament. Once activated, FLS are suggested to acquire a myofibroblast-like phenotype that drives fibrogenesis through excessive extracellular matrix component deposition and an enhanced contractile function. These cells play a crucial role in the pathogenesis of chronic inflammatory diseases, such as rheumatoid arthritis. Therefore, Human FLS are an important model to understand articular function in degenerative and inflammatory joint diseases[1-3].

iXCells Biotechnologies provides high quality Human Fibroblast-Like Synoviocytes- Osteoarthritis (HFLS-OA), which are isolated from osteoarthritic human synovial membrane and cryopreserved at P2, with ≥0.5 million cells in each vial. HFLS-OA express fibronectin and vimentin and are characterized by their spindle-shaped morphology. They are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi and can be further expanded for no more than 3 passages in Synoviocyte Growth Medium (Cat# MD-0056) under the conditions suggested by iXCells Biotechnologies.

Product Description

Fibroblasts are malleable cell types, capable of altering its function and physiology or even transforming into a new cell type, based on its location within the body. Gastric fibroblasts, isolated from the stomach, are one of the mesenchymal cells derived from the embryonic mesoderm. Human Gastric Fibroblasts are responsible for secretion of extracellular matrix in connective tissues and play a key role in ulcer wound healing [1,2]. Several studies have shown that gastric fibroblasts can regulate epithelial regeneration by influencing epithelial cell proliferation and differentiation. In contrast to their role in wound healing, human gastric fibroblasts have shown to be a key contributor in the development and progression of gastric carcinomas [1,3]. Therefore, human gastric fibroblasts are a useful model for examining the underlying mechanisms that cause gastric carcinoma and may provide novel insights for developing new cancer drug therapies.

iXCells Biotechnologies provides high quality Human Gastric Fibroblasts isolated from the stomach of healthy donors. Each vial contains ≥0.5 million cells cryopreserved at P1 and are delivered frozen. iXCell’s Human Gastric Fibroblasts are characterized by immunofluorescence using antibodies specific to fibronectin and vimentin. These cells are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi. They can be further expanded in Fibroblast Growth Medium (Cat# MD-0011) for no more than 3 passages under the condition suggested by iXCells biotechnologies.

Product Description

Fibroblasts, one of the most abundant cell types, are morphologically and functionally heterogeneous [1]. Human pulmonary fibroblasts (HPF) play an important role in maintaining the structural integrity of connective tissue and in the synthesis of extracellular matrix proteins such as collagens and glycoproteins [2]. Under pathological conditions, such as tissue injury, HPF are stimulated to take part in the process of airway inflammation and airway remodeling which might directly or indirectly lead to angiogenesis, idiopathic fibrosis or even cancer [3]. Idiopathic pulmonary fibrosis (IPF) is a rare, progressive illness of the respiratory system, characterized by replacement of healthy lung tissue with altered extracellular matrix and alveolar architecture is destroyed. IPF is believed to be the result of an aberrant wound healing process involving abnormal fibroblasts activation [4].

iXCells Biotechnologies provides high quality HPF, which are isolated from lung tissue of IPF patients. It provides a robust model for in vitro studies of IPF as well as other fibroblasts related diseases. HPF from iXCells Biotechnologies are cryopreserved at P2, with >0.5 million cells in each vial. HPF express Fibronectin and Vimentin. They are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi and can further expand for 10 population doublings in Fibroblast Growth Medium (Cat# MD-0011) under the condition suggested by iXCells Biotechnologies.

Product Description

Glomerular podocytes in kidney are highly specialized visceral epithelial cells with a complex cytoarchitecture. These cells feature finger-like projections called “foot process” that interlock with each other, creating tiny gaps known as filtration slits that play a crucial role in filtering blood within the glomerulus [1,2]. Podocytes are involved in a variety of glomerular functions, including glomerular basement membrane turnover, maintenance of filtration barrier, support of the capillary tuft, regulation of glomerular filtration and immunological functions. Injury to podocytes may lead to proteinuria, a hallmark of most glomerular diseases and chronic kidney disease. Disruptions of podocyte architecture resulting in the retraction of foot processes and proteinuria are common features in the progression of acquired glomerular disease [3,4].

iXCells Biotechnologies offers high quality primary Human Podocytes isolated from human kidney obtained by following IRB protocol and cryopreserved at P2, with ≥ 0.5 million viable cells in each vial. These podocytes have been characterized by the proliferation, and the expression of cell type specific markers, including CD2AP, Nephrin, NPHS2, Podocin, Synaptopodin (Figure 1, 2). Human Podocytes are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi. These cells can be maintained in Podocyte Culture Medium (Cat# MD-0120) and expanded for 2~3 passages under conditions suggested by iXCells Biotechnologies. Further expansion may decrease podocyte particular traits such as foot processes and expression of slit diaphragm proteins.

Product Description

The adrenal cortex makes up the perimeter of the adrenal gland and plays an essential role in regulating homeostasis in the body through the secretion of corticosteroid and androgen hormones[1] . The secreted steroids arise from three zones that form the adrenal gland, which provide the framework of the adrenal cortex. The cells in the cortex stem from the mesoderm and form three concentric zones named the zona glomerulosa, zona fasciculate, and zona reticularis[2] . Studies have shown that there is extensive interaction between the cortical and medulla regions of the adrenal gland, with cortical cells being found within the adrenal medulla and chromaffin cells within the adrenal cortex. The close contact of these two cell types implies intercellular exchange and allows for further studies concerning paracrine signaling between the two adrenal endocrine systems[3] . Human adrenal cortex also harbours a mesenchymal stem cell-like population[4]. Adrenal cortical cells can be used for research of hormonal regulation, steroidogenesis and regenerative therapy in adrenal insufficiency.

iXCells Biotechnologies provides high quality HAdCC, which were isolated from adult normal adrenal cortical tissue. HAdCC were cryopreserved at passage one (P1), with >0.5 million cells in each vial. They are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi and can further expand for no more than 3 passages in Adrenal Cortical Cell Growth Medium (Cat# MD-0117) under the condition suggested by iXCells Biotechnologies.

Figure 1. Phase contrast images of HAdCC. The primary cells predominantly comprised adrenocortical cells with polygonal appearance and light-refractive lipid core. Some adrenal cortical fibroblasts-like cells were also noticed in early passages. The images were taken at magnification of 10× and 20×.

Figure 2. Fluorescent immunostaining images of HAdCC. Immunocytochemistry staining was performed using antibodies against 17 beta hydroxysteroid dehydrogenase 7 (HSD17β7) and vimentin. The cells immunoreactive with HSD17β7 are steroid hormone producing cells. Nuclei were counterstained with DAPI.

Product Description

The liver is a vital organ in mammals, and hepatocytes, the major cell type in liver, play critical roles in protein synthesis, detoxification of various metabolites, transformation of carbohydrates, and the production of biochemical necessary for digestion [1]. Primary hepatocyte culture is an important model for in vitro studies in drug development, including hepatotoxicity, drug transport, hepatitis virus infection, hepatic drug metabolism and hepatobiliary excretion [2].

iXCells Biotechnologies provides high quality human primary hepatocytes isolated from whole liver of human organ donors and cryopreserved on the day of isolation with ≥5 million viable cells in each vial. iXCells offers human hepatocytes in plateable & suspension categories and includes cells from both healthy and diseased tissue. Our cryoplateable primary human hepatocytes (CPHH) reach and maintain a confluent monolayer (>90% confluency) for 5-7 days (Fig 1), express general hepatocyte marker albumin, and show accumulation of lipid droplets (Fig 2). Each lot of human hepatocytes is QC tested & validated including key Cytochromes P450 (CYPs) enzyme activity and gene expression induction. They are negative for HIV-1/2, HBV, HCV, mycoplasma, bacteria, yeast, and fungi.

Figure 1. Phase contrast images of CPHH. CPHH were plated in a 6-well culture dish precoated with Collagen type I and cultured for 5 days following iXCells protocol. CPHH reached and maintained monolayer with >90% confluency. Hepatocytes exhibited hexagonal shape with single or double nuclei and characteristic “chicken wire” like bile canaliculus formation between hepatocytes.

Product Description

Mesenchymal stem cells (MSC) derived from human placenta are a well-characterized population of adult stem cells. Number of researchers have shown that adult MSCs have a broad therapeutic potential due to their capability to renew and differentiate into various lineages of mature cells that produce fat, cartilage, bone, tendon, and muscle when cultured under specific permissive conditions [1]. In addition, placenta derived MSC show differentiation capacity toward both osteo-blasts and adipocytes thus serving as a great model for studying the molecular basis of differentiation [2]. These properties, in combination with their developmental plasticity, have generated tremendous interest in regenerative medicine to replace damaged tissues. These findings have spurred the development of MSC-based therapies for treating wide range of non-skeletal diseases [3].

iXCells Biotechnologies provides placental MSC isolated from different layers of human placenta, including decidua (Cat# 10HU-170), chorionic villi (Cat# 10HU-171), and chorionic plate (Cat# 10HU-172). Each vial contains ≥ 0.5 million cells. These cells are expanded in Mesenchymal Stem Cell Medium (Cat# MD-0037) and then cryopreserved at passage 2. The human placental MSC express typical mesenchymal cell surface markers, such as CD105, CD73, and CD90 and are negative for hematopoietic markers including CD34, CD45 and endothelial cell marker CD31 [Figure 1, 2]. These cells can further be differentiated into adipocytes using Adipocyte Differentiation Medium (Cat# MD-0005) and into osteoblasts using Osteogenic Differentiation Medium (Cat# MD-0006) [Figure 3 and Figure 4]. Human placental MSC are negative for mycoplasma, bacteria, yeast, and fungi and can be expanded for no more than 3 passages in iXCells’ Mesenchymal Stem Cell Medium.

Product Description

Fibroblasts and myofibroblasts are believed to be the key effector cells in renal fibrogenesis responsible for the synthesis and deposition of extracellular matrix components [1]. Although fibroblasts are histologically visible in normal kidneys, there are relatively few of them and proximal tubular epithelial cells predominate. In progressive disease, however, the interstitium becomes filled with myofibroblasts [2]. Fibroblasts are considered the primary matrix-producing cells in the kidney and hence they are clinically relevant as principal mediators of renal fibrosis associated with progressive renal failure [3].

iXCells Biotechnologies provides high quality Human Renal Fibroblasts, which are isolated from adult human kidney tissue and cryopreserved at P1, with >0.5 million cells in each vial. Human Renal Fibroblasts express fibronectin and are negative for HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi. They can further expand no more than 3 passages in Fibroblast Growth Medium (Cat# MD-0011) under the condition suggested by iXCells Biotechnologies.
